intranasal delivery comes up often in conversation and rarely with the context attached. Here we lay out the basics in order, then work through the practical considerations.

Published research covers ischemic stroke, traumatic brain injury, cognitive impairment, optic nerve conditions and attention-related measures. Much of the human evidence comes from small trials conducted in one country, which limits how far the results generalize. Animal models supply the larger share of the data, and effects seen in rodents do not transfer automatically to people. Reviews have noted that methodological reporting is often incomplete, making it difficult to pool results or compare treatment schedules across studies.
Pharmacokinetic accounts emphasize rapid breakdown. After intravenous dosing the intact peptide disappears from blood within minutes, and nasal delivery produces low but measurable concentrations. Metabolites rather than the parent molecule may account for part of the observed activity, although the relative contribution is unresolved. Dosing in the literature varies widely and no optimal schedule has been agreed. These gaps are regularly cited as a reason the findings have not produced broad clinical adoption beyond the original research setting.
Proposed mechanisms center on neurotrophic signaling rather than on classical melanocortin receptor activation. Rodent experiments have reported shifts in the expression of brain-derived neurotrophic factor and nerve growth factor after administration, together with changes in the associated receptor systems. Several authors argue that the peptide acts largely through its degradation products and their interaction with peptidergic pathways, but this remains a hypothesis rather than a settled finding. No single molecular target has been identified in a way that the field broadly accepts.
Circulation time for the peptide is short because peptidases cleave it readily. The Pro-Gly-Pro tail is thought to slow breakdown compared with the bare ACTH fragment, but the gain appears modest. Absorption after intranasal dosing is limited, and only a fraction of a dose is expected to reach the central nervous system. Laboratory concentrations therefore sit well above levels achieved systemically, a gap that complicates translation from bench findings to clinical claims.
The mechanisms attributed to semax are inferred from animal and cell studies rather than traced to one confirmed target. The most frequently cited pathway involves increased expression of brain-derived neurotrophic factor and nerve growth factor in hippocampal and cortical tissue. Some work points to engagement of melanocortin receptors, particularly MC4, which the parent ACTH fragment can activate. Effects on dopaminergic and serotonergic signalling have also been reported. No single account explains all observed results, and the relative weight of each pathway remains unsettled.

Claims about enhanced focus, memory, or mood in healthy people rest mostly on anecdotal reports and community discussion rather than on controlled data. It remains unclear whether any cognitive benefit observed in patients recovering from brain injury would extend to uninjured users. Dose-response relationships, long-term safety, and interactions with other drugs are not well characterized in the peer-reviewed literature. Questions about optimal route of administration and treatment duration likewise remain open.
Semax binds to melanocortin receptors and is thought to influence neuronal survival and plasticity rather than to act through the adrenal axis. Laboratory work has shown increased expression of brain-derived neurotrophic factor and nerve growth factor in treated tissue. Changes in c-Fos, a marker of neuronal activation, have also been reported. Because the peptide is rapidly degraded by peptidases, its effects are generally attributed to downstream signaling cascades rather than to sustained receptor occupancy.
== Neurology == Research into the notochord has played a key role in understanding the development of the central nervous system. By transplanting and expressing a second notochord near the dorsal neural tube, 180 degrees opposite of the normal notochord location, one can induce the formation of motor neurons in the dorsal tube. Motor neuron formation generally occurs in the ventral neural tube, while the dorsal tube generally forms sensory cells. The notochord secretes a protein called sonic hedgehog (SHH), a key morphogen regulating organogenesis and having a critical role in signaling the development of motor neurons. The secretion of SHH by the notochord establishes the ventral pole of the dorsal-ventral axis in the developing embryo.
In 2009, Chipotle released an app for the iPhone that allows users to find nearby Chipotle locations, place an order, and prepay with a credit card. In 2013, Chipotle released an Android app that allows users to locate nearby Chipotle locations, place an order, prepay with a credit or gift card, and access favorites and recent orders. In late 2021, the company added a non-food item to the menu—cilantro soap. According to published reports, the soap sold out the day after its release. The soap was produced after an August 2021 Instagram post of what was thought to be a mock photo of the product. It was an effort to engage fans and haters of the herb that, "plays into a larger trend of turning digital moments into real life experiences," according to Chris Brandt, Chipotle's Chief Marketing Officer. Chipotle uses green marketing and its "Food with Integrity" platform to boost its brand image.
A myxoma (New Latin from Greek muxa 'mucus') is a myxoid tumor of primitive connective tissue. It is most commonly found in the heart (and is the most common primary tumor of the heart in adults) but can also occur in other locations.
At the active site, a substrate binds to an enzyme to induce a chemical reaction. Substrates, transition states, and products can bind to the active site, as well as any competitive inhibitors. For example, in the context of protein function, the binding of calcium to troponin in muscle cells can induce a conformational change in troponin. This allows for tropomyosin to expose the actin-myosin binding site to which the myosin head binds to form a cross-bridge and induce a muscle contraction. In the context of the blood, an example of competitive binding is carbon monoxide which competes with oxygen for the active site on heme. Carbon monoxide's high affinity may outcompete oxygen in the presence of low oxygen concentration. In these circumstances, the binding of carbon monoxide induces a conformation change that discourages heme from binding to oxygen, resulting in carbon monoxide poisoning.
Counter-Strike has over 20 years of competitive play beginning with the original Counter-Strike. The first major tournament was hosted in 2001 at the Cyberathlete Professional League which, along with World Cyber Games and Electronic Sports World Cup, were among the largest tournaments for the Counter-Strike series up to 2007. Since 2013, the Valve-sponsored Counter-Strike: Global Offensive Major Championships have become the most prestigious tournaments in the franchise's history, featuring prize pools of around $1,000,000.

== Biosimilar insulin == A biosimilar is a biological medicine that is highly similar to an already approved reference biologic in terms of structure, biological activity, efficacy, and safety. These medicines are large, complex molecules produced through biotechnology in living systems, such as microorganisms, plant cells, or animal cells. Due to differences in the manufacturing process, biosimilars cannot be exact copies of reference biologics but must demonstrate high similarity through extensive structural and functional analysis. Manufacturers are required to show that a biosimilar has no clinically meaningful differences from its reference product regarding safety, purity, and potency, which is assessed through pharmacokinetic (PK) and pharmacodynamic (PD) studies, immunogenicity evaluations, and, if necessary, additional clinical studies. Biosimilars can only be developed and marketed once the patent on the original reference biologic has expired, allowing for competition and increased availability of biologic therapies. The expiration of patents for first-generation insulin analogs has facilitated the development of biosimilar insulins, offering potential to improve global insulin access. Despite the essential role of insulin, approximately half of individuals who require it do not have access due to high costs and limited availability. This issue is particularly pronounced in low-income countries, where economic factors can restrict the use of biologic treatments such as insulin.
== Statements by Combatants' Presidents and Prime ministers == On 7 August, state-controlled Rossiya TV aired Abkhaz separatist leader Sergei Bagapsh, who said at a meeting of the Abkhaz National Security Council: "I have spoken to the president of South Ossetia. It has more or less stabilized now. A battalion from the North Caucasus District has entered the area." The phone conversation between Bagapsh and Kokoity had taken place on late 6 August. By the evening of 7 August, Bagapsh had ordered the Abkhaz armed forces to raise combat readiness. Pro-Georgian president of South Ossetia Dmitry Sanakoyev told Russian journalist Dmitry Steshin by 8 August 2008 that neither he nor the Georgian government needed the war and cited multi-billion Georgian investment into the development of South Ossetia as a proof that Georgia was not preparing the war. Prime Minister Lado Gurgenidze said on 8 August that South Ossetian separatists launched a deliberate attack on the Georgian civilians after Saakashvili's announcement of ceasefire and that 100-vehicle armed convoy had entered through the Roki Tunnel from Russia before the midnight. Gurgenidze said that the Georgian government had notified the command of the Russian peacekeepers that Georgia was forced to respond to the attack. On 11 August 2008, Georgian president Mikheil Saakashvili wrote that the war was provoked by the Kremlin and while Georgia was showing restraint, Russia escalated the conflict in South Ossetia.
== Pathophysiology == The exact cause of venous ulcers is not certain. A common denominator is generally venous stasis, which may be caused by chronic venous insufficiency, and/or congestive heart failure. Venous stasis causes the pressure in veins to increase. The body needs the pressure gradient between arteries and veins for the heart to pump blood forward through arteries and into veins. When venous hypertension exists, arteries no longer have significantly higher pressure than veins, and blood is not pumped as effectively into or out of the area. Venous hypertension may also stretch veins and allow blood proteins to leak into the extravascular space, isolating extracellular matrix (ECM) molecules and growth factors, preventing them from helping to heal the wound. Leakage of fibrinogen from veins as well as deficiencies in fibrinolysis may also cause fibrin to build up around the vessels, preventing oxygen and nutrients from reaching cells. Venous insufficiency may also cause white blood cells (leukocytes) to accumulate in small blood vessels, releasing inflammatory factors and reactive oxygen species (ROS, free radicals) and further contributing to chronic wound formation. Buildup of white blood cells in small blood vessels may also plug the vessels, further contributing to ischemia. This blockage of blood vessels by leukocytes may be responsible for the "no reflow phenomenon", in which ischemic tissue is never fully reperfused. Allowing blood to flow back into the limb, for example by elevating it, is necessary but also contributes to reperfusion injury.
Accurate and uniform temperature in a refrigerator plays a key role in ensuring the life of vaccines, reagents and other biologicals. Research has shown that minor variances in temperature such as those in a household refrigerator can compromise the effectiveness of your biologicals, risking up to thousands of dollars in valuable contents. Vaccines are also compromised through improper use of the door gasket to feed cables from data loggers and thermometers, allowing excess warm air in, and cold air out of the refrigerator or freezer. Over time this causes the compressor to work a longer duty cycle and eventually leads to failure. This can be remedied by using probe access ports, found on most clinical refrigerators and freezer. These are easy to open up and drastically reduce air intake and loss from inside the units.
A separate consequence is that the freshly milled particles carry high surface energy and acquire substantial triboelectric charge by repeated particle–wall and particle–particle contact during milling and downstream handling. The resulting electrostatic forces drive agglomeration, complicate blending uniformity, and — in dry powder inhaler formulations — cause drift in fine-particle fraction during storage as the powder relaxes toward thermodynamic equilibrium. To mitigate these effects, micronized actives intended for high-sensitivity applications are commonly subjected to a post-milling conditioning step, in which the powder is exposed to controlled humidity or a solvent vapour for hours to days to allow surface relaxation, partial recrystallization, and reduction of stored mechanical energy before formulation. Mechanical micronization is generally unsuitable for biologic APIs such as proteins, because the high-energy stresses of jet milling can cause denaturation; spray drying with stabilizing excipients is the route typically used to produce inhalable dry powders of biologics.

The main proposal is modulation of neurotrophic factors such as brain-derived neurotrophic factor, supported largely by animal experiments. Receptor-level targets have not been firmly established. Most reviews describe the mechanism as only partially characterized.
The compound is registered as a medicine in Russia, so clinical work has concentrated there. Trials elsewhere are few and generally small. This geographic concentration is a recognized limitation in evidence reviews.
Some fraction may reach the central nervous system through olfactory pathways, and this is often cited as the rationale for the nasal route. The size of that fraction in humans is not well quantified. Blood concentrations after nasal dosing are low, which complicates measurement.
It consists mainly of animal experiments and small clinical reports, with much of the clinical material published in Russian-language journals. Large independent trials are scarce. Separating reliable effects from chance findings is consequently difficult.
