A certificate of analysis that pairs a chromatogram with a mass spectrum is more informative than a purity figure alone. === Mass spectrometry analysis === Mass spectrometry is currently the best method to adequately compare pairs of protein samples. The two main procedures to perform this task are using isotope-coded affinity tags (ICAT) and stable isotopic amino acids in cell culture (SILAC). In the ICAT procedure samples are labeled individually after isolation with mass-coded reagents that modify cysteine residues. In SILAC, cells are cultured separately in the presence of different isotopically labeled amino acids for several cell divisions allowing cellular proteins to incorporate the label. Mass spectrometry is subsequently used to identify phosphoserine, phosphothreonine, and phosphotyrosine-containing peptides.
== Early life and education == Robert S. Swanson was born in Brooklyn, New York, in 1947 to Arthur J. Swanson and Arline Baker Swanson. Arthur Swanson was an airplane electrical maintenance crew leader, and worked in shifts. According to Swanson, he was taught from an early age that his generation would do better than the last generation of his family. It was because of this that his family wanted him to be the first to obtain a college degree. His family was particularly interested in the Massachusetts Institute of Technology (MIT). Much to his family's pride, Swanson was accepted into MIT in 1965. Even though he was majoring in chemistry, he realized later during his undergraduate education that he preferred working with people, rather than in research. What follows is an excerpt from a 1996 interview that describes how he came to this realization: "At the end of my junior year, I... got a summer job working for a chemical company... One of the things I discovered was that I enjoyed people more than things. So I said, 'Gee, this probably isn't going to be what I'd want to do all my life,'". As a result, Swanson petitioned MIT to be able to take the first year's courses at the Alfred P. Sloan School of Management for a master's degree, and they allowed him to do so. Thanks to the graduate courses he took, he realized that he was particularly interested in two things: organizational development, and the commercialization of innovative ideas. He graduated from MIT in 1970, with an undergraduate degree in chemistry and a Master of Science degree in management.
== Lead optimization phase == The objective of this drug discovery phase is to synthesize lead compounds, new analogs with improved potency, reduced off-target activities, and physiochemical/metabolic properties suggestive of reasonable in vivo pharmacokinetics. This optimization is accomplished through chemical modification of the hit structure, with modifications chosen by employing knowledge of the structure–activity relationship (SAR) as well as structure-based design if structural information about the target is available. Lead optimization is concerned with experimental testing and confirmation of the compound based on animal efficacy models and ADMET (in vitro and in situ) tools that may be followed by target identification and target validation.

=== Opioid use disorder === Buprenorphine is used to treat people with opioid use disorder. In the U.S., the combination formulation of buprenorphine/naloxone is generally prescribed to deter injection, since naloxone, an opioid antagonist, is believed to cause acute withdrawal if the formulation is crushed and injected. Taken orally, the naloxone is still capable of blocking the rewarding effects of Buprenorphine. Especially given its combination with certain medications that can reduce the metabolism of naloxone. Before starting buprenorphine, individuals with opioid dependence are generally advised to wait after their last dose of opioid, often 24–72 hours, because if taken too soon buprenorphine can displace other opioids bound to the receptors and precipitate an acute withdrawal. The dose of buprenorphine is then adjusted until symptoms improve, and individuals remain on a maintenance dose, often 8–16 mg. Because withdrawal is uncomfortable and a deterrent for many patients, users have called for different means of treatment initiation. The Bernese method, also known as microdose induction, was described in 2016, where very small doses of buprenorphine (0.2 to 0.5 mg) are given while patients are still using street opioids, with medicine levels slowly titrated upward without precipitating withdrawal. This method has been used by some providers as of the 2020s. Later he performs onstage at the Roxy Theatre wearing a shirt featuring the image of a pill labeled "Rorer 714". Quaaludes are mentioned in the 1983 film Scarface, when Al Pacino's character Tony Montana says, "Another Quaalude... she'll love me again." Small white pills referred to as "ludes" appear along with several other illicit drugs in the 1983 comedy-drama The Big Chill. == Terms == Analyte – the substance to be separated during chromatography. It is also normally what is needed from the mixture. Analytical chromatography – the use of chromatography to determine the existence and possibly also the concentration of analyte(s) in a sample. Bonded phase – a stationary phase that is covalently bonded to the support particles or to the inside wall of the column tubing. Chromatogram – the visual output of the chromatograph. In the case of an optimal separation, different peaks or patterns on the chromatogram correspond to different components of the separated mixture. Plotted on the x-axis is the retention time and plotted on the y-axis a signal (for example obtained by a spectrophotometer, mass spectrometer, or a variety of other detectors) corresponding to the response created by the analytes exiting the system. In the case of an optimal system the signal is proportional to the concentration of the specific analyte separated. Chromatograph – an instrument that enables a sophisticated separation, e.g. gas chromatographic or liquid chromatographic separation. Chromatography – a physical method of separation that distributes components to separate between two phases, one stationary (stationary phase), the other (the mobile phase) moving in a definite direction. == Research == AAS, alone and in combination with progestogens, have been studied as potential male hormonal contraceptives. Dual AAS and progestins such as trestolone and dimethandrolone undecanoate have also been studied as male contraceptives, with the latter under active investigation as of 2018. Topical androgens have been used and studied in the treatment of cellulite in women. Topical androstanolone on the abdomen has been found to significantly decrease subcutaneous abdominal fat in women, and hence may be useful for improving body silhouette. However, men and hyperandrogenic women have higher amounts of abdominal fat than healthy women, and androgens have been found to increase abdominal fat in postmenopausal women and transgender men as well.

Many of these methods can be automated for high throughput production but care must be taken to avoid conditions of synthesis or extraction that result in a denatured protein which, since it no longer recognizes its binding partner, renders the array useless.
