The peptide is prone to several degradation pathways. Oxidation of the methionine residue produces a sulfoxide that elutes close to the parent peak in many chromatographic systems. Hydrolysis of peptide bonds and deamidation of susceptible residues in related sequences also reduce purity over time. Lyophilised material kept dry at minus twenty degrees Celsius and shielded from light is the most stable form commonly described in laboratory practice.
Material sold for laboratory use varies widely in stated purity and documentation. A certificate of analysis should list the analytical method, the column and detector used, and the observed purity value. Independent verification by an outside laboratory is the practical way to confirm identity when documentation is absent or internally inconsistent. Regulatory status differs by country, and a product legal in one jurisdiction may be unapproved or controlled in another.
Semax is a synthetic seven-amino-acid peptide whose sequence extends the ACTH(4-10) fragment with a C-terminal proline-glycine-proline tripeptide. The commonly cited sequence is Met-Glu-His-Phe-Pro-Gly-Pro, giving a molecular formula near C37H51N9O10S and a molecular weight close to 813.9 g/mol. It belongs to the broader class of synthetic ACTH fragments studied for central nervous system effects rather than for adrenal steroid stimulation. In practice the material appears as a lyophilized white powder for laboratory work or as a dilute saline solution in clinical settings.
Development is attributed to researchers at the Institute of Molecular Genetics in Moscow during the early 1980s, building on earlier Soviet work with ACTH fragments. Russian regulatory approval followed for intranasal use, and the compound has remained commercially available there for decades. Most published human data originate from Russian and, later, some Eastern European clinical reports, which are not always accessible in English translation. Outside that region the material is generally handled as a research chemical rather than a licensed medicine.

Published studies are dominated by animal models of stroke, ischemia, and cognitive impairment, with a smaller number of human trials conducted in Russia. Many of the human reports are small, single-center, and published in Russian-language journals, which limits independent scrutiny. Outcome measures vary between studies and often rely on clinician-rated scales rather than objective biomarkers. Systematic reviews have noted the methodological weaknesses and called for larger, preregistered trials before firm conclusions can be drawn.
Claims about enhanced focus, memory, or mood in healthy people rest mostly on anecdotal reports and community discussion rather than on controlled data. It remains unclear whether any cognitive benefit observed in patients recovering from brain injury would extend to uninjured users. Dose-response relationships, long-term safety, and interactions with other drugs are not well characterized in the peer-reviewed literature. Questions about optimal route of administration and treatment duration likewise remain open.
Semax binds to melanocortin receptors and is thought to influence neuronal survival and plasticity rather than to act through the adrenal axis. Laboratory work has shown increased expression of brain-derived neurotrophic factor and nerve growth factor in treated tissue. Changes in c-Fos, a marker of neuronal activation, have also been reported. Because the peptide is rapidly degraded by peptidases, its effects are generally attributed to downstream signaling cascades rather than to sustained receptor occupancy.
The compound was developed in the 1980s at the Institute of Molecular Genetics in Moscow, where it emerged from research on short ACTH fragments and their effects on the central nervous system. Russian pharmaceutical listings describe it as a nootropic and neuroprotective agent, most often formulated as nasal drops. It is not a marketed medicine in the United States or the European Union, and no pharmacopoeial monograph covers it. Consequently, most published clinical experience with the substance originates from a small number of research centres, mainly in Russia and neighbouring countries.
Pharmacological accounts link semax to melanocortin signalling and to modulation of neurotrophic factor expression, particularly brain-derived neurotrophic factor and nerve growth factor. Much of this evidence comes from rodent studies using intranasal delivery, a route chosen because it allows peptides to reach the central nervous system with limited systemic exposure. Whether the same mechanisms operate in humans at comparable magnitude remains an open question. The precise receptor or receptors responsible for the reported behavioural and neuroprotective effects have not been conclusively identified.
Clinical reports describe use in ischaemic stroke, transient ischaemic attack, optic nerve conditions, and cognitive complaints, but most of these studies are small and were conducted in a single region. Systematic reviews have generally described the evidence base as limited in size and variable in methodological quality. Randomised controlled data suitable for international regulatory assessment are scarce. As a result, major treatment guidelines outside Russia do not include the peptide, and interest in it remains largely research-driven rather than routine clinical.
Semax is supplied as a lyophilized powder that appears white to off-white. It dissolves readily in water, phosphate-buffered saline, and other aqueous media, which simplifies preparation of working solutions for laboratory use. The nasal products registered in Russia are dilute aqueous solutions, typically around 0.1 percent peptide by weight. Organic solvents are rarely necessary and can complicate handling. Because the peptide is hygroscopic, weighing should be performed quickly and with minimal exposure to ambient humidity.
Solid material is normally kept at minus 20 degrees Celsius in a sealed, desiccated container. Reconstituted solutions are less stable and are usually divided into single-use aliquots before freezing. Repeated freeze-thaw cycles are avoided because they promote aggregation and loss of activity. Light exposure is minimized by using amber glassware or foil wrapping. Published stability data for this peptide are sparse, so recommended storage conditions rest mainly on general practice for short synthetic peptides rather than on dedicated study.
The Miller–Urey experiment, or Miller experiment, was an experiment in chemical synthesis carried out in 1952 that simulated the conditions thought at the time to be present in the atmosphere of the early, prebiotic Earth. It is seen as one of the first successful experiments demonstrating the synthesis of organic compounds from inorganic constituents in an origin of life scenario. The experiment used methane (CH4), ammonia (NH3), hydrogen (H2), in ratio 2:2:1, and water (H2O). Applying an electric arc (simulating lightning) resulted in the production of amino acids. It is regarded as a groundbreaking experiment, and the classic experiment investigating the origin of life (abiogenesis). It was performed in 1952 by Stanley Miller, supervised by Nobel laureate Harold Urey at the University of Chicago, and published the following year. At the time, it supported Alexander Oparin's and J. B. S. Haldane's hypothesis that the conditions on the primitive Earth favored chemical reactions that synthesized complex organic compounds from simpler inorganic precursors. After Miller's death in 2007, scientists examining sealed vials preserved from the original experiments showed that more amino acids were produced in the original experiment than Miller reported with paper chromatography. While evidence suggests that Earth's prebiotic atmosphere might have typically had a composition different from the gas used in the Miller experiment, prebiotic experiments continue to produce racemic mixtures of simple-to-complex organic compounds, including amino acids, under varying conditions.
Diabetes – general term referring to all states characterized by hyperglycemia. It can be of the following types: Type 1 diabetes – autoimmune-mediated destruction of insulin-producing β-cells in the pancreas, resulting in absolute insulin deficiency Type 2 diabetes – either inadequate insulin production by the β-cells or insulin resistance or both because of reasons not completely understood. there is correlation with diet, with sedentary lifestyle, with obesity, with age and with metabolic syndrome. Causality has been demonstrated in multiple model organisms including mice and monkeys; importantly, non-obese people do get Type 2 diabetes due to diet, sedentary lifestyle and unknown risk factors, though this may not be a causal relationship. it is likely that there is genetic susceptibility to develop Type 2 diabetes under certain environmental conditions Other types of impaired glucose tolerance (see Diabetes) Insulinoma – a tumor of beta cells producing excess insulin or reactive hypoglycemia. Metabolic syndrome – a poorly understood condition first called syndrome X by Gerald Reaven. It is not clear whether the syndrome has a single, treatable cause, or is the result of body changes leading to type 2 diabetes. It is characterized by elevated blood pressure, dyslipidemia (disturbances in blood cholesterol forms and other blood lipids), and increased waist circumference (at least in populations in much of the developed world).
=== Lawsuit === In November 2012, GlaxoSmithKline was ordered by a Rennes appeals court to pay Frenchman Didier Jambart 197,000 euros ($255,824); Jambart had taken ropinirole from 2003 to 2010 and exhibited risky hypersexual behavior and gambled excessively until stopping the medication. This behavior displayed is characteristic of Dopamine Dysregulation Syndrome.

It is hypothesized that the various introductions led to multiple genotypes which are adapted to either oaks or conifers. Californian specimens of A. phalloides frequently display yellowish to mustard-yellow caps, a coloration that can closely resemble the edible species Amanita velosa and Amanita calyptroderma, both of which are commonly foraged and consumed in California. This deceptive appearance is the result of environmental homoplasy with Asian cousin Amanita subjunquillea: the fungus expresses a phylogenetically conserved pigment palette (derived from the shikimate pathway) under the particular microclimatic conditions of coastal California oak woodlands. Prolonged high humidity from summer fog drip, followed by intense sunlight and oxidative stress once the fog burns off, upregulates polyphenolic compounds and causes oxidative bleaching of the originally greener or olive tones, producing the characteristic yellowish hues. Such similarity poses a significant risk of fatal misidentification for foragers, as the highly toxic death cap may be mistaken for these otherwise edible look-alikes. A. phalloides were conveyed to new countries across the Southern Hemisphere with the importation of hardwoods and conifers in the late twentieth century. Introduced oaks appear to have been the vector to Australia and South America; populations under oaks have been recorded from Melbourne, Canberra (where two people died in January 2012, of four who were poisoned), Adelaide, and further observed by citizen scientists in Beechworth, Sydney and Albury.
=== Hypnotic === Chloral hydrate has not been approved by the FDA in the United States nor the EMA in the European Union for any medical indication and is on the FDA list of unapproved drugs that are still prescribed by clinicians. Usage of the drug as a sedative or hypnotic may carry some risk given the lack of clinical trials. However, chloral hydrate products, licensed for short-term management of severe insomnia, are available in the United Kingdom. Chloral hydrate was voluntarily removed from the market by all manufacturers in the United States in 2012. Prior to that, chloral hydrate may have been sold as a "legacy" or "grandfathered" drug; that is, a drug that existed prior to the time certain FDA regulations took effect and therefore, some pharmaceutical companies have argued, has never required FDA approval. New drugs did not have to be approved for safety until Congress passed the Federal Food, Drug, and Cosmetic Act (the "FD&C Act") in 1938. Further, a new drug did not have to be proven effective until 1962, when Congress amended the Act. Manufacturers contend that such "legacy drugs", by virtue of the fact that they have been prescribed for decades, have gained a history of safety and efficacy. Chloral hydrate was used for the short-term treatment of insomnia and as a sedative before minor medical or dental treatment. It was largely displaced in the mid-20th century by barbiturates and subsequently by benzodiazepines.
The latest development in the medical treatment of acromegaly is the use of growth hormone receptor antagonists. The only available member of this family is pegvisomant (Somavert). By blocking the action of the endogenous growth hormone molecules, this compound is able to control the disease activity of acromegaly in virtually everyone with acromegaly. Pegvisomant has to be administered subcutaneously by daily injections. Combinations of long-acting somatostatin analogues and weekly injections of pegvisomant seem to be equally effective as daily injections of pegvisomant. Paltusotine (Palsonify) was approved for medical use in the United States in September 2025.
Since the oxychlorides are asymmetrical, and they should have increasingly large electric dipole moments going down the group, they should become less volatile in the order TcO3Cl > ReO3Cl > BhO3Cl: this was experimentally confirmed in 2000 by measuring the enthalpies of adsorption of these three compounds. The values are for TcO3Cl and ReO3Cl are −51 kJ/mol and −61 kJ/mol respectively; the experimental value for BhO3Cl is −77.8 kJ/mol, very close to the theoretically expected value of −78.5 kJ/mol.

Expert Tornike Sharashenidze wrote in November 2017 that "the annexation of Crimea and the beginning of the war in Donbass discredited Russia, which allowed the ‘guilt’ of the 2008 August war to be lifted from Georgia."
== Factor structure == Although Cohen et al. (1983) originally argued the PSS to be a unidimensional measure of perceived stress, the research community generally views the 14- and 10-item forms as two dimensional. The predominant forms consist of positively and negatively phrased items. Under exploratory factor analysis the negatively phrased items have been found to load onto a second factor separate from the positively phrased items, giving the appearance of a method effect. That is, a significant portion of the variability in the responses to a subset of the items is a product of how the items are phrased. However, inspection of the items suggests a substantive difference in the positively phrased items versus the negatively phrased items with the negatively phrased items on their face characterizing perceived helplessness while the positively phrased items may be characterizing perceived self-efficacy. Some support for this conceptualization has emerged. For example, Hewitt et al. (1992) found that the perceived helplessness items tended to predict depression in both men and women while the positively phrased items tended to predict depression in women only. The pattern of differential prediction of depression between genders tends to indicate the negatively phrased items are tapping something substantive rather than a method effect.
=== Neuroprotective effects === Animal experiments with diabetic rats have shown that 1-methylnicotinamide positively effects degenerative changes in the brain, allowing cognitive performance to be maintained longer. It also prevents depressive behavior with efficacy comparable to the common antidepressant fluoxetine. This effect is attributed to the reduction of neuroinflammation, pro-inflammatory cytokines (IL-6, TNF-α), and increased expression of BDNF (brain-derived neurotrophic factor), a protein supporting neuron survival and growth. The neuroprotective effects of 1-MNA involve shielding against neurotoxins, amyloid-beta plaques in the brain, neuroinflammatory responses, and neuronal apoptosis. It has been shown to improve memory deficits and cognitive functions, suggesting potential for treating neurodegenerative disorders.
== Variant tags == This tag is also known as the C-terminal TAP tag because an N-terminal version is also available. However, the method to be described assumes the use of a C-terminal tag, although the principle behind the method is still the same.

Reversed-phase high-performance liquid chromatography is the standard method, with detection in the ultraviolet range. Peak area percentage yields a purity figure for the main component. Mass spectrometry is normally run alongside to confirm molecular identity.
Dry lyophilised powder is generally kept at minus twenty degrees Celsius, protected from light and moisture. Repeated freeze-thaw cycles are avoided because they encourage aggregation and moisture uptake. Solutions are less stable than the solid form and degrade faster at room temperature.
A certificate documents what the supplier measured, not what an independent party confirmed. Methods, instruments and acceptance criteria can differ between suppliers. Third-party testing is the usual way to resolve discrepancies.
It is a short synthetic peptide built from seven amino acids: methionine, glutamic acid, histidine, phenylalanine and three prolines. The sequence derives from the 4-10 fragment of adrenocorticotropic hormone with an added proline-glycine-proline tail. No plant or animal extract is involved; the material is produced by solid-phase peptide synthesis.
